What is the Significance of the Kingdom of Babylon in Biblical History?

What ancient kingdom/empire represented Babylon?  Was it Babel, under Nimrod or was it Babylon, under Nebuchadnezzar?  This gets a little confusing.  Also, in Revelation, Babylon appears again.  What is that about?
